چکیده

Methionine sulfoxide reductases A and B (MsrA and MsrB) have been known to be thioredoxin (Trx)-dependent enzymes that catalyze the reduction of methionine sulfoxide in a stereospecific manner. This work reports that glutaredoxin, another major thiol-disulfide oxidoreductase, can serve as a reductant for both MsrA and MsrB. Glutaredoxins efficiently reduced 1-Cys MsrA lacking a resolving Cys, which is not reducible by Trx. Glutaredoxins also reduced 3-Cys MsrA containing two resolving Cys. The glutaredoxin-dependent activity of the 3-Cys MsrA was comparable with the Trx-dependent activity. The kinetic data suggest that 1-Cys MsrA is more efficiently reduced by glutaredoxin than 3-Cys form. Also, glutaredoxins could function as a reductant for 1-Cys MsrB lacking a resolving Cys as previously reported. In contrast to the previous report, 2-Cys MsrB containing a resolving Cys was reducible by the glutaredoxins. Collectively, this study demonstrates that glutaredoxins reduce MsrAs and MsrBs with or without resolving Cys.

Regeneration mechanisms of Arabidopsis thaliana methionine sulfoxide reductases B by glutaredoxins and thioredoxins.

Methionine oxidation leads to the formation of S- and R-diastereomers of methionine sulfoxide (MetSO), which are reduced back to methionine by methionine sulfoxide reductases (MSRs) A and B, respectively. MSRBs are classified in two groups depending on the conservation of one or two redox-active Cys; 2-Cys MSRBs possess a catalytic Cys-reducing MetSO and a resolving Cys, allowing regeneration b...

Atypical thioredoxins in poplar: the glutathione-dependent thioredoxin-like 2.1 supports the activity of target enzymes possessing a single redox active cysteine.

Plant thioredoxins (Trxs) constitute a complex family of thiol oxidoreductases generally sharing a WCGPC active site sequence. Some recently identified plant Trxs (Clot, Trx-like1 and -2, Trx-lilium1, -2, and -3) display atypical active site sequences with altered residues between the two conserved cysteines. The transcript expression patterns, subcellular localizations, and biochemical propert...

Thionein can serve as a reducing agent for the methionine sulfoxide reductases.

It has been generally accepted, primarily from studies on methionine sulfoxide reductase (Msr) A, that the biological reducing agent for the members of the Msr family is reduced thioredoxin (Trx), although high levels of DTT can be used as the reductant in vitro. Preliminary experiments using both human recombinant MsrB2 (hMsrB2) and MsrB3 (hMsrB3) showed that although DTT can function in vitro...

Evidence for the Dimerization-Mediated Catalysis of Methionine Sulfoxide Reductase A from Clostridium oremlandii

Clostridium oremlandii MsrA (CoMsrA) is a natively selenocysteine-containing methionine-S-sulfoxide reductase and classified into a 1-Cys type MsrA. CoMsrA exists as a monomer in solution. Herein, we report evidence that CoMsrA can undergo homodimerization during catalysis. The monomeric CoMsrA dimerizes in the presence of its substrate methionine sulfoxide via an intermolecular disulfide bond ...

Methionine sulfoxide reductase A and a dietary supplement S-methyl-L-cysteine prevent Parkinson's-like symptoms.

Parkinson's disease (PD), a common neurodegenerative disease, is caused by loss of dopaminergic neurons in the substantia nigra. Although the underlying cause of the neuronal loss is unknown, oxidative stress is thought to play a major role in the pathogenesis of PD. The amino acid methionine is readily oxidized to methionine sulfoxide, and its reduction is catalyzed by a family of enzymes call...
